FORMULATION OF SELF-NANOEMULSIFYING DRUG DELIVERY SYSTEM (SNEDDS) HYDROCHLOROTIAZIDE USING LONG CHAIN TRIGYCERIDE (LCT) AS OIL PHASE

Abstract

Hydrochlorotiazide (C7H8CIN3O4S2) is an effective thiazide diuretic drug for lowering blood pressure, classified in BCS class IV because it has low solubility and permeability. The purpose of this study was to determine the best formula of SNEDDS HCT using Long Chain Triglyceride oil. Nanoemulsions (o/w) are made using hydrochlorotiazide as the active substance, LCT oil as the oil phase, Tween 80 and PEG 400 as surfactants and cosurfactants. The oil selected for SNEDDS hydrochlorotiazide preparation is isopropyl myristate. The ratio produced by the optimal formula of SNEDDS between isopropyl myristate : Tween 80 : PEG 400 is 11.11% : 66.67% : 22.22% with HCT as much as 25 mg / mL SNEDDS. SNEDDS HCT showed a transmittance % of 97.454%, emulsification time of 22.61 seconds, particle size of 14.6 nm, zeta potential of -31.2mV, viscosity of 3 cpss, pH 6.63, and good stability during storage. Dissolution testing showed increased percent dissolution of SNEDDS HCT. The SNEDDS HCT functional group test corresponds to the functional group characteristics of the HCT, which indicates the presence of the drug in the formulation.
